The auto exposure from phyphox will adapt the camrea settings to achieve this and this brightness level is mostly unaffected by the post-processing of your phone. The relative luminance is then almost entirely based on the camera settings (especially ISO and exposure time) that are required to achieve this brightness. However, the auto exposure takes a moment to settle, so you should wait a moment before taking relative luminance measure and observe if the preview image settles in a medium gray brightness.
